Bu çalışmada, organofosfatlı pestisit olan methyl parathion ve dichlorvos’un eritrositler
üzerinde serbest radikal üreterek oksidatif stres meydana getirdiği ve hücresel antioksidan
savunma sistemini değiştirdiği gösterilmiştir. Bu tez çalışmasında in vitro koşullarda farklı
dozlarda methyl parathion ve dichlorvos (1,10,100 µM) ve methyl parathion/dichlorvos
vitamin C (VC; 10µM)/vitamin E (VE; 30µM) kombinasyonunun insan eritrositlerindeki
malondialdehit (MDA) seviyesi ile süperoksit dismutaz (SOD), katalaz (CAT) ve glutatyon
peroksidaz (GPx) enzim aktiviteleri üzerine olan etkileri incelenmiştir. Eritrositler farklı
uygulamalarda (sadece methyl parathion, sadece dichlorvos, yalnızca vitaminler, methyl
parathion+vitaminler ve dichlorvos+vitaminler) 37 ºC’de 60 dk inkübe edilmişlerdir ve
MDA seviyesi ile SOD, CAT ve GPx enzim aktiviteleri araştırılmıştır. Methyl paration ve
dichlorvos tek başlarına uygulandıklarında eritrositlerde MDA seviyesini arttırdıkları; SOD,
CAT ve GPx aktivitelerinde ise azalma meydana getirdikleri gözlenmiştir (P<0,05). VCuygulamalı, VE-uygulamalı ve VC+VE-uygulamalı eritrositler uygulama yapılmayan
kontrol hücreleri ile karşılaştırıldığında istatistiksel olarak bir farklılık gözlenmemiştir.
Methyl parathion/dichlorvos+VC ve methyl parathion/dichlorvos+VE ile muamele edilen
gruplarda methyl parathion ve dichlorvos’un lipit peroksidasyonu ve antioksidan enzim
aktivitelerinde meydana getirdikleri değişikler engellenmişlerdir. Ancak bu yararlı etki
sadece methyl parathion ve dichlorvos’un düşük uygulama dozlarında (1 ve 10µM)
görülmüş ve VC+VE kombinasyonunun bu vitaminlerin tek başlarına kullanılmalarından
daha fazla koruyucu etkiye sahip olduğu gözlenmiştir. Elde edilen bu sonuçlar vitaminlerin
plazmada bulunan konsantrasyonlarının, methyl parathion ve dichlorvos’un genelde pestisit
olarak kullanılan yüksek dozlarının (100 µM) eritrositlerde oluşturdukları zararlı etkileri
üzerine koruyu olmadığını göstermiştir. In this study, methyl parathion and dichlorvos are organophosphate (OP) pesticide that have
been shown to induce oxidative stress in erythrocytes through the generation of free radicals
and alteration of the celluler antioxidant defense system. We examinated the effect of several
different doses of methyl parathion and dichlorvos (1,10,100µM) or methyl parathion or
dichlorvos in combination vitamin C (VC; 10µM) or vitamin E (VE; 30µM), on the levels of
malondialdehyde (MDA), superoxide dismutase (SOD), catalase (CAT) and glutathione
peroxidase (GPx) activities in human erythrocytes in vitro. Erythrocytes were incubated
under various treatment conditions (methyl parathion alone, dichlorvos alone, vitamins
alone, methyl parathion plus vitamins, and Dichlorvos plus vitamins) at 37 ºC for 60 min,
and the levels of MDA, SOD, CAT and GPx activities, were determined. Treatment with
methyl paration or dichlorvos alone increased the levels of MDA, and decreased SOD, CAT
and GPx activities in erythrocytes (P<0,05). There were no statistical different among VCtreated, VE-treated, or VC+VE-terated erythrocytes, as compared with nontreated control
cells. Treatment of cells with methyl parathion or dichlorvos+VC, methyl parathion or
dichlorvos+VE, or a combination of all three agents prevented Methyl parathion or
dichlorvos-induced changes in antioxidant enzyme activity and lipid peroxidation. However,
this effect was seen only at low concentrations of methyl parathion or dichlorvos (1 and
10µM), and combination of VC+VE had a more protective effect than VC or VE alone.
These results indicated that the presence of vitamins at concentrations that are similar to the
levels found in plasma have no effect on methyl parathion or dichlorvos-induced toxicity in
erythrocytes at a concentration of methyl parathion or dichlorvos (100 µM) that are typically
used in pesticides.
